CB-011 is an off-the-shelf, allogeneic anti-BCMA CAR-T cell therapy developed by Caribou Biosciences for the treatment of relapsed or refractory multiple myeloma. CB-011 is manufactured from healthy donor T cells and engineered ex vivo using Caribou’s CRISPR-based platform (Cas12a chRDNA) to incorporate an immune cloaking strategy, including knockout of the B2Mgene and insertion of a B2M-HLA-E fusion protein, designed to reduce host immune recognition and rejection of the allogeneic CAR-T cells. The therapy is intended to provide a readily available, single-dose treatment option without the manufacturing delays associated with autologous CAR-T therapies. CB-011 is being evaluated in the Phase 1 CaMMouflage clinical trial and has received FDA Fast Track and Orphan Drug designations.
