CRISPR On/Off: Modelling and Predicting On-/Off-Targets for CRISPR/Cas9 gRNAs

Accurate prediction of gRNA on-target efficiency with minimum off-targets is critical for the CRISPR workflow. In this presentation, the speaker will present two tools, CRIPSRon and CRISPRoff, which can be used for finding gRNAs with highest efficiency and to carry out an off-target assessment. The presentation will be in two parts:

  1. Focusing on CRISPR/Cas9 design and introducing the basic methodological concepts from deep learning to energy modelling of CRISPR/Cas9 as well as the introduction of a completely new tool CRISPRroots for on/off-target quality analysis post the CRISPR editing.
  2. The online available versions of the CRISPRon/off tools will be introduced.

Jan Gorodkin, is a bioinformatician and professor at the Center for non-coding RNA in Technology and Health, in the Department of Veterinary and Animal Sciences, at University of Copenhagen, Denmark. He received his PhD from the Technical University of Denmark and has since then worked with several topics within computational RNA biology, genome and transcriptome analyses. Over the years, his group has published numerous computational tools for sequence and RNA structure analysis. Within recent years, his group took up work on computational methods for CRISPR analysis with outset in computational methods for RNA structure and interaction analysis.
